The PhD project will focus on the characterization of P. falciparum-induced knobs on the red blood cell surface in a time resolved manner, using super-resolution microscopy, including STED and MINFLUX, and various electron microscopic techniques, including cryo-electron tomography. All required instruments are accessible on the Heidelberg University campus Im Neuenheimer Feld.

Red blood cells infected with the human malaria parasite Plasmodium falciparum acquire adhesive properties and, as a result, sequester in the deep vascular bed to avoid splenic clearance mechanisms. Cytoadhesion of infected erythrocytes is mediated by families of adhesins that are presented on protrusions, termed knobs. Knobs not only present the adhesins; knobs also anchor the adhesins to the membrane skeleton of the red blood cell; and they cause stiffening of the membrane. Cells devoid of knobs do not cytoadhere. A critical building block of knobs is the knob-associated histidine-rich protein (KAHRP), which, although being highly unstructured, can interact with different components of the erythrocyte actin/spectrin network and parasite factors. However, the exact time course of the interactions are not known yet.

The objective of the PhD project is to study knob assembly in a time-resolved manner and identify critical interactions of KAHRP with parasite and host cell factors. A library of KAHRP mutants is available.

Qualified candidate should have a Master’s degree from an internationally accredited institution of higher education. Technical skills in super resolution microscopy and/or electron microscopy are desirable. Interest in interdisciplinary collaborations is mandatory. Applications from all fields of natural sciences are welcome as are candidates with a migration background. We also encourage scientists from Eastern European countries, including Ukraine, and Russia to apply.
